I wonder how much a car/moto will cost? Staffed by Greeks and fares starting at $100/person sound nice!

Great news. That was the missing link in my planning. The Ferry must be high enough to take standard roof Sprinters?
Thank for sharing.

Seems as though the May 10, 2012 start-up date came and went without a ferry. As far as I can tell, the Nissos Rodos is still in Greece.

Also, the website www.panaferry.com (http://www.panaferry.com) is not responding.
However, this person seems to have been able to get some information, with a new start up date of 5/31/2012 - date that does not seem likely given that the ship is still in Greece as of last AIS reading:

Update 5/23 - now Panamerican Seaways is blaming the governments for not issuing permits. Of course, the Nissos Rodos is still in Greece. See link above. This is looking more and more like a scam, as I can find no indications that Hellenic Seaways has any intention of leasing or otherwise deploying the Nissos Rodos to this route. There is no indication that Panamerican Seaways is related to Hellenic Seaways, and the link provided by Panamerican Seaways is only to the Hellenic Seaways' ship site - not their media center. This could be an exercise in misdirection - attempting to create the impression of a relationship between the two where none exists.
Update 5/30 - there are now reports of the company refusing to honor refund requests. Surprise surprise.
Update 6/26 - The ferry allegedly headed to Panama has moved - but it is not going anywhere - the move was from the shipyard
